Learn more.Mastitis is the single most common and costly disease of dairy cattle worldwide, mainly caused by intra-mammary infection (IMI). The bactotype Mastitis HP3 PCR Kit is a highly sensitive and specific solution for the detection of DNA from three major mastitis-causing pathogens – Streptococcus agalactiae (S. agalactiae), Mycoplasma bovis (M. bovis ) and Staphylococcus aureus (S. aureus) – in samples from ruminant milk. Principle The bactotype Mastitis HP3 PCR Kit detects the most important cow-associated and highly contagious bacteria S. agalactiae, M. bovis and S. aureus in milk samples, by amplifying specific regions of the pathogen genome through polymerase chain reaction (PCR). In real-time PCR, the amplified product is detected using fluorescent dyes. These are usually linked to oligonucleotide probes that bind specifically to the amplified product. Monitoring the fluorescence intensities during the PCR run (i.e., in real time) allows the detection of the accumulating product without the need to re-open the reaction tubes afterwards. The bactotype Mastitis HP3 PCR Kit contains all of the necessary reagents for the detection of S. agalactiae, M. bovis and S. aureus DNA, including a positive control, a negative control and an internal control.
Procedure The kit contains a ready-to-use mix with all of the necessary reagents for the detection of DNA from S. agalactiae, M. bovis and S. aureus and the required positive and negative controls. DNA is extracted from the starting material (e.g., quarter milk samples, pool milk samples or tank milk samples) using a suitable sample preparation kit. QIAGEN recommends using the DNeasy Mastitis Mini Kit (192) for manual DNA isolation using silica membrane/column technology or the MagAttract Mastitis Kit (384) for automated DNA isolation using magnetic bead technology. The extracted DNA is used for real-time PCR, which is run on a thermal cycler. Applications The bactotype Mastitis HP3 PCR Kit is for detection of DNA from S. agalactiae, M. bovis and S. aureus in ruminant milk samples. |
